產(chǎn)品介紹 High Mobility Group Box-1 (HMGB1) is a cytokine implicated in the pathogenesis of rheumatoid arthritis (RA) and other inflammatory diseases. The cholinergic anti-inflammatory pathway, a vagus nerve dependent mechanism, inhibits HMGB1 release in experimental disease models

Function:
DNA binding proteins that associates with chromatin and has the ability to bend DNA. Binds preferentially single-stranded DNA. Involved in V(D)J recombination by acting as a cofactor of the RAG complex. Acts by stimulating cleavage and RAG protein binding at the 23 bp spacer of conserved recombination signal sequences (RSS). Heparin-binding protein that has a role in the extension of neurite-type cytoplasmic processes in developing cells.

近來的研究表明稱之為高遷移率族蛋白B-1(HMG-B1)的核內(nèi)結(jié)構(gòu)蛋白在核外表達(dá)時(shí)是一種有效的早期炎癥介質(zhì)。
高遷移性B1組蛋白(HMGB1): 是一種核結(jié)合蛋白,在DNA重組、修復(fù)、復(fù)制和基因轉(zhuǎn)錄中起作用。HMGB1也是巨噬細(xì)胞分泌的一種介質(zhì)。
此外,高遷移性B1組蛋白亦被受刺激的巨噬細(xì)胞或單核細(xì)胞主動(dòng)分泌。在這一主動(dòng)分泌過程中,HMGB1首先經(jīng)乙?;⒂珊藘?nèi)轉(zhuǎn)移至溶酶體內(nèi),繼而在ATP和溶血磷脂膽堿兩種分泌信號(hào)指導(dǎo)下轉(zhuǎn)移至細(xì)胞外。由壞死細(xì)胞被動(dòng)釋放的HMGB1和炎癥細(xì)胞主動(dòng)分泌的HMGB1存在分子上的差異。胞外的HMGB1可作為細(xì)胞因子參與信號(hào)傳導(dǎo),因?yàn)樗瓤勺R(shí)別Toll 樣受體(TLR)家族的一些成員,又能與識(shí)別晚期糖基化終末產(chǎn)物受體(RAGE)相作用。
HMGB1能啟動(dòng)炎癥反應(yīng),包括產(chǎn)生多種細(xì)胞因子、對(duì)某些干細(xì)胞產(chǎn)生趨化作用、誘導(dǎo)血管粘附分子、削弱腸上皮細(xì)胞的功能等。

Secondary Antibody (white blue line): Goat anti-rabbit IgG-FITC Dilution: 1μg /test. Protocol The cells were fixed with 70% ethanol (Overnight at 4℃) and then permeabilized with 90% ice-cold methanol for 30 min on ice. Cells stained with Primary Antibody for 30 min at room temperature. The cells were then incubated in 1 X PBS/2%BSA/10% goat serum to block non-specific protein-protein interactions followed by the antibody for 15 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.

Secondary Antibody : Goat anti-rabbit IgG-AF488 Dilution: 1μg /test. Protocol The cells were fixed with 4% PFA (10min at room temperature)and then permeabilized with 90% ice-cold methanol for 20 min at-20℃. The cells were then incubated in 5%BSA to block non-specific protein-protein interactions for 30 min at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.
Rat splenocytes stained with Anti- HMGB1 Polyclonal Antibody, A488 Conjugated (bs-0664R-A488) at 1:50.
